Assessing Genetic Variation and Spread of Phalaris minor Resistant to ACCase Inhibiting Herbicides in Iran Planta Daninha
GHEREKHLOO,J.; ALCÁNTARA-DE LA CRUZ,R.; OSUNA,M.D.; SOHRABI,S.; PRADO,R..
ABSTRACT: Littleseed canarygrass (Phalaris minor) is the second most serious and problematic grass weed in wheat fields in Iran, and has developed resistance to ACCase inhibiting herbicides. Inter-simple sequence repeat (ISSR) analysis was used to assess genetic variation between and within ACCase inhibitor-resistant and susceptible P. minor populations in Iran and to determine the origin of resistance and its dispersal. Sixteen P. minor populations from different regions in Iran were analysed using seven primers. Genetic relationships generated using UPGMA analysis indicated the presence of more than one genotype among the herbicide resistant populations. The results indicated that the high genetic similarity and physical proximity among the resistant P....

Assessment of Genetic Fidelity of in vitro Raised Plants in Swertia chirayita through ISSR, RAPD analysis and Peroxidase Profiling during Organogenesis BABT
Sharma,Vikas; Belwal,Nidhi; Kamal,Barkha; Dobriyal,Anoop Kumar; Jadon,Vikash Singh.
ABSTRACT An efficient in vitro regeneration protocol for medicinally important herb Swertia chirayita was developed and the genetic fidelity was assessed using RAPD and ISSR markers. The best shoot regeneration was observed on MS basal supplemented with 1.0 mg/L Benzyl amino purine (BAP) in combination with Indole-3-acetic acid (IAA) (0.5 mg/L) that resulted in the increase by multiplication rate (7.65) with an average of 33.33 numbers of shoots and average shoot length of 2.70 cm. It was further enhanced by the addition of adenine sulfate (0.007%) that resulted in an average of 42 shoots per clum with 4.13 cm of average shoot length and the increase in multiplication fold to 9.75 that further resulted in the reduced use of other cytokinins and auxins. The...

Assessment of somaclonal variation in indirect morphogenesis-derived plants of Arracacia xanthorrhiza PAB
Vitamvas,Jan; Viehmannova,Iva; Cepkova,Petra Hlasna; Mrhalova,Hana; Eliasova,Katerina.
Abstract: The objective of this work was to induce and detect somaclonal variation in arracacha (Arracacia xanthorrhiza) plants regenerated via indirect morphogenesis, in order to evaluate the potential of this technique to produce new genotypes for breeding purposes of this crop. Calli were induced from petiole segments on Murashige & Skoog (MS) medium supplied with 0.1 mg L-1 2,4-dichlorophenoxyacetic acid. The regeneration of plants via indirect morphogenesis was carried out on half-strength MS medium without plant growth regulators. Fifteen randomly chosen plants were subjected to flow cytometry and “inter-simple sequence repeat” (ISSR) analysis. Ploidy level remained stable in all tested regenerants (2n=4x=44), with no changes in the genome....

BSA and molecular markers screening for salt stress tolerant mutant of Petunia obtained in in vitro culture Ciência Rural
Krupa-Małkiewicz,Marcelina; Bienias,Anna.
ABSTRACT: In this study, we performed BSA to identify genetic markers linked to salt tolerance. We tested the genetic diversity among four bulked DNA samples of EMS induced mutant clones and one bulked DNA sample of non-mutated clone of Petunia for salt tolerance in in vitro callus cultures using RAPD and ISSR markers. Out of the 36 RAPD and 16 ISSR primers identified, 25 and 13 were effectively used to amplify genomic DNA of all the five bulked samples, respectively. In total, 114 RAPD amplifications products were obtained, of which 28% were polymorphic and 2% were genotype-specific bands. Out of the 64 ISSR amplification products obtained, 51% were polymorphic and 1% was genotype-specific bands. Results of this study indicated the existence of two...

Clonal diversity and conservation genetics of the medicinal plant Carapichea ipecacuanha (Rubiaceae) Genet. Mol. Biol.
Oliveira,Luiz Orlando de; Venturini,Bruna Afonso; Rossi,Ana Aparecida Bandini; Hastenreiter,Saulo Santos.
The roots of the understorey shrub Carapichea ipecacuanha (ipecac) have medicinal properties, and the uprooting of wild plants has supplied most of the world demand for this species. Although under severe population decline, C. ipecacuanha lacks legal protection. In the wild, the aerial stems of ipecac clump together to form clusters with well-defined borders. Cluster size may range from several to hundreds of aerial stems. To investigate the extent of clonality among aerial stems in ipecac clusters, we sampled 50 wild clusters (a total of 291 aerial stems) and screened them with 89 inter-simple sequence repeat (ISSR) markers. The 291 aerial stems were grouped into 42 putative clones. The clonal groups generally consisted of aerial stems from the same...

Determination of genetic variation in Rhodiola crenulata from the Hengduan Mountains Region, China using inter-simple sequence repeats Genet. Mol. Biol.
Lei,Yidong; Gao,Hong; Tsering,Tashi; Shi,Suhua; Zhong,Yang.
The plant Rhodiola crenulata is a perennial herbaceous species distributed in the plateau region of southwestern China, especially the Hengduan Mountains region. It has been one of the most important traditional herbal remedies in Tibet for more than one thousand years, but the accelerated and uncontrolled collection of this plant since the 1980s has lead to deforestation. We used inter-simple sequence repeats (ISSR) to assess levels of genetic variation in R. crenulata from nine diverse natural populations in eastern Tibet and northern Yunnan, the first time such a study has been carried out. The 12 primers we used were able to detect 184 polymorphic loc. Analysis of molecular variance (AMOVA) indicated that species level genetic diversity was relatively...

Development of sequence characterized amplified region markers from intersimple sequence repeat fingerprints for the molecular detection of toxic phytoplankton Alexandrium catenella (Dinophyceae) and pseudo-Nitzschia pseudodelicatissima (Bacillariophyceae) from French coastal waters ArchiMer
Bornet, Benjamin; Antoine, Elisabeth; Francoise, Sylvaine; Marcaillou-le Baut, Claire.
Harmful algal blooms are a serious threat to shellfish farming and human health all over the world. The monitoring of harmful algae in coastal waters originally involved morphological identification through microscopic examinations, which was often difficult unless performed by specialists and even then often did not permit identification of toxic species. More recently, specific molecular markers have been used to identify specific phytoplankton species or strains. Here we report on the use of the intersimple sequence repeat (ISSR) technique to develop specific sequence characterized amplified region markers (SCAR) and to identify with these tools two toxic species in French coastal waters, the diatom Pseudo-nitzschia pseudodelicatissima (Hasle) Hasle and...

Efficiency of RAPD and ISSR markers in assessing genetic variation in Arthrocnemum macrostachyum (Chenopodiaceae) BABT
Saleh,Basel.
The goal of this study was to investigate the molecular characterization of A. macrostachyum genotypes grown in the Western coast of Syria using RAPD and ISSR techniques. PCR amplifications with 20 RAPD primers gave an average of 9.25 selected markers/ primer, with a maximum of 17 (OPG05) and a minimum of four (OPA02). Percentage of polymorphic bands ranged from 40 to 100% according to RAPD primers tested. Among the 185 selected bands, 160 (84.96%) were polymorphic. The amplification with seven ISSR primers generated 88 bands, and 80 (90.91%) were polymorphic. (CA)8RG and (AG)8GTG ISSR primers tested in this study yielded highly informative patterns. RAPD and ISSR fragment sizes ranged from 0.2-3 kb. Based on this study, the use of RAPD and ISSR...

Genetic diversity and structure of Atta robusta (Hymenoptera, Formicidae, Attini), an endangered species endemic to the restinga ecoregion Genet. Mol. Biol.
Reis,Evelyze Pinheiro dos; Salomão,Tânia Maria Fernandes; Campos,Lucio Antonio de Oliveira; Tavares,Mara Garcia.
The genetic diversity and structure of the ant Atta robusta were assessed by ISSR (inter-simple sequence repeats) in 72 colonies collected from 10 localities in the Brazilian states of Espírito Santo (48 colonies) and Rio de Janeiro (24 colonies). The ISSR pattern included 67 bands, 51 of them (76.1%) polymorphic. Analysis of molecular variance (AMOVA) revealed a high level (57.4%) of inter-population variation, which suggested a high degree of genetic structure that was confirmed by UPGMA (unweighted pair-group method using an arithmetic average) cluster analysis. The significant correlation between genetic and geographic distances (r = 0.64, p < 0.05) indicated isolation that reflected the distance between locations. Overall, the populations were...

Fajardo,Cristiane Gouvêa; Costa,Daniel Ferreira da; Chagas,Kyvia Pontes Teixeira das; Vieira,Fábio de Almeida.
ABSTRACT The continuing fragmentation of forests has been a threat to the maintenance of genetic resources. Genetic diversity is fundamental to the survival of species in natural environments in the long term, as well as being the basis for genetic improvement. The objective of this study was to evaluate the genetic diversity in natural populations of Hancornia speciosa and to contribute to the development of conservation strategies. We sampled 105 individuals of H. speciosa, distributed in seven populations. The ISSR (Inter-Simple Sequence Repeat) markers provided 70 loci, of which 81% were polymorphic. The mean genetic diversity of Nei (h) was 0.19, and the Shannon index (I) was 0.27. The h and I diversity indices ranged respectively from 0.16 to 0.24 in...

Chagas,Kyvia Pontes Teixeira das; Freire,Ageu da Silva Monteiro; Pinheiro,Luciana Gomes; Fajardo,Cristiane Gouvêa; Vieira,Fábio de Almeida.
Abstract This study aimed to estimate the genetic diversity of three Elaeis guineensis populations in Rio Grande do Norte state, as well as to verify the decreases in effective population size. The population with the highest polymorphism was MAT (Mata) with+ 57 loci (72%), followed by RIA (Riacho) with 54 loci (68%) and HOR (Horta) with 34 loci (43.03%). The RIA population was shown to be the most genetically diverse, with Nei (h = 0.28) and Shannon (I = 0.41) diversity indices. There was high genetic differentiation among populations (AMOVA, analysis of molecular variance = 42%), which was separated into three distinct genetic groups according to a Bayesian analysis. There was a significant population decrease (P < 0.05) for the HOR population in the...

Genetic diversity of populations of the dioecious Myrsine coriacea (Primulaceae) in the Atlantic Forest Acta Botanica
Paschoa,Roberta Pena da; Christ,Jheniffer Abeldt; Valente,Cecília Silva; Ferreira,Marcia Flores da Silva; Miranda,Fábio Demolinari de; Garbin,Mário Luís; Carrijo,Tatiana Tavares.
ABSTRACT Although a species’ sexual system may influence the genetic diversity of its populations in their natural environment, there have been few such studies involving indigenous species of the Atlantic Forest. Here we study Myrsine coriacea, a dioecious tree widely used in reforestation programs despite a lack of information about its natural interpopulation genetic variation. To address this knowledge gap, intra- and interpopulation genetic diversity were measured for male and female individuals of ten natural populations using ISSR markers. Greater intrapopulation genetic diversity indicated interpopulation gene flow, regardless of isolation and distance between populations. Multivariate analyses detected significant differences in genetic diversity...

Genetic evidence of multiple reproductive strategies in a microendemic and threatened cactus (Cactaceae: Discocactus Pfeiff) in Bahia, Brazil Acta Botanica
Jesus,Izabela Santos Dias de; Conceição,Leila Patricio; Schnadelbach,Alessandra Selbach; Assis,José Geraldo de Aquino; Carvalho,Maria Luiza Silveira de.
ABSTRACT Discocactus zehntneri subsp. petr-halfari, an endangered taxon, is represented by a single population in an anthropized area of Bahia, Brazil, where it is suffering due to extreme extractivism. Thus, information about this cactus, such as its reproductive patterns, is urgently needed to support conservation strategies. A population genetics approach was used to determine if this subspecies has a preferential pattern of reproduction. We sampled 18 individuals, both with and without connection to parental plants, from five clumps and assessed their diversity and genetic structure using five ISSR markers. The results revealed two clumps that are genetically supported by the presence of genetically equal individuals. The other three groups presented...

Genetic relationship in Coffea species and parentage determination of interspecific hybrids using ISSR (Inter- Simple Sequence Repeat) markers Genet. Mol. Biol.
Ruas,Paulo M.; Ruas,Claudete F.; Rampim,Leandro; Carvalho,Valdemar P.; Ruas,Eduardo A.; Sera,Tumoru.
Inter-simple sequence repeat (ISSR) markers were used to evaluate genetic divergence among eight Coffea species and to identify the parentage of six interspecific hybrids. A total of 14 primers which contained different simple sequence repeats (SSR) were used as single primers or combined in pairs and tested for PCR amplifications. Two hundred and thirty highly reproducible fragments were amplified, which were then used to estimate the genetic similarity and to cluster the Coffea species and hybrids. High levels of interspecific genetic variation were revealed. The dinucleotide motif (GA)9T combined with other di- tri- and tetra-nucleotides produced a greater number of DNA fragments, mostly polymorphics, suggesting a high frequency of the poly GA...

Genetic variation in cultivated Rheum tanguticum populations Genet. Mol. Biol.
Hu,Yanping; Xie,Xiaolong; Wang,Li; Zhang,Huaigang; Yang,Jian; Li,Yi.
To examine whether cultivation reduced genetic variation in the important Chinese medicinal plant Rheum tanguticum, the levels and distribution of genetic variation were investigated using ISSR markers. Fifty-eight R. tanguticum individuals from five cultivated populations were studied. Thirteen primers were used and a total of 320 DNA bands were scored. High levels of genetic diversity were detected in cultivated R. tanguticum (PPB = 82.19, H = 0.2498, H B = 0.3231, I = 0.3812) and could be explained by the outcrossing system, as well as long-lived and human-mediated seed exchanges. Analysis of molecular variance (AMOVA) showed that more genetic variation was found within populations (76.1%) than among them (23.9%). This was supported by the coefficient...

Hakki,Erdogan E; Kayis,Seyit A; Pinarkara,Emine; Sag,Ayla.
Cannabis sativa L. is a multiple-use plant that provides raw material for the production of seed oil, natural fiber for textiles, automotive and pulp industries. It has also been used in insulating boards, ropes, varnishes, animal feed, and as medicinal agents. Cannabis has potential to be used for phytoremediation: however, its cultivation is strictly controlled due to its psychoactive nature and usage in producing drugs such as marijuana, and hashish. In this study, psychoactive type Cannabis samples, which were seized from 23 different locations of Turkey, and nine hemp type Cannabis accessions, as well as an unknown accession were used. Our interest was to identify the genetic relatedness of the seized samples and to separate drug and hemp type plants....
